Sus antecedentes:

  • Las proteínacinasas son enzimas cruciales, pero ~ 10% se clasifican como pseudokinasas debido a la falta de residuos catalíticos.
  • Las pseudocinasas se consideraron en gran medida catalizadoras, lo que limita sus funciones funcionales conocidas.

Objetivo del estudio:

  • Para investigar la actividad funcional de la pseudokinasa conservada, la selenoproteína-O (SelO).
  • Explorar las actividades enzimáticas potenciales más allá de la función de la quinasa canónica para las pseudokinasas.

Principales métodos:

  • Cristalografía de rayos X para determinar la base estructural de la actividad de SelO.
  • Ensayos in vitro para demostrar la transferencia de AMP a sustratos de proteínas.
  • Estudios de localización celular y análisis del papel de SelO en la respuesta al estrés oxidativo.

Principales resultados:

  • SelO cataliza la transferencia de AMP del ATP a los residuos de Ser, Thr y Tyr en sustratos de proteínas, un proceso llamado AMPilación.
  • La estructura cristalina revela un pliegue similar a la quinasa con un sitio de unión al ATP alterado, lo que explica el mecanismo catalítico.
  • SelO se localiza en las mitocondrias y las proteínas AMPylates que regulan la homeostasis redox, lo que demuestra su papel en la defensa celular.

Conclusiones:

  • Las pseudokinasas, ejemplificadas por SelO, poseen nuevas actividades de transferasa como la AMPilación.
