  • Comprender la influencia de la estereoregularidad en la UCST es clave para el diseño de materiales.

Objetivo del estudio:

  • Investigar la influencia de la estereoregularidad del polipéptido en el comportamiento de la temperatura crítica superior de la solución (UCST).
  • Para comparar la separación de fase y la agregación de polipéptidos homoquirales y racémicos basados en citrulina.
  • Explorar la base estructural y la biodegradabilidad de estos polímeros térmicamente sensibles.

Principales métodos:

  • Síntesis y caracterización de variantes de poliornitina y cocitrulina (POC) con una estereoregularidad variable.
  • Medición de las temperaturas de separación de fases (Tps) mediante análisis térmico.
  • Análisis estructural mediante dicroísmo circular (CD) y dispersión de rayos X de ángulo pequeño (SAXS).

Principales resultados:

  • La poli (l-ornithine-co-l-citrulline) homoquírea (PlOC) y la poli (d-ornithine-co-d-citrulline) (PdOC) exhibieron Tps más altos que la poli (dl-ornithine-co-dl-citrulline) racémica (PdlOC).
  • PdlOC mostró separación de fase líquido-coacervado, mientras que PlOC y PdOC formaron agregados similares a los sólidos.
  • Los POC homoquirales adoptaron estructuras alfa helicoidales y se ensamblaron en redes hexagonales, impulsadas por las interacciones del grupo ureido.

Conclusiones:

  • La estereorregularidad dicta críticamente el comportamiento UCST, los modos de separación de fase y la organización estructural en polipéptidos basados en citrulina.
  • El plegamiento helicoidal y el ensamblaje hexagonal observados en POCs homoquirales se atribuyen a las interacciones pendentes del grupo ureido.
  • Las estructuras separadas por fases influyen en la biodegradabilidad, destacando el potencial de estos polímeros de tipo UCST para aplicaciones de biomateriales sintonizables.
