    Sus antecedentes:

    • Los péptidos cortos son bloques de construcción versátiles para el autoensamblaje en biomateriales funcionales de hidrogel.
    • Los conjuntos de péptidos supramoleculares ofrecen un enfoque atractivo para aplicaciones de ingeniería de tejidos neurales.

    Objetivo del estudio:

    • Para reportar una nueva clase de péptidos cortos de cinco residuos que forman hidrogeles con estructuras de nanofibra.
    • Caracterizar las propiedades mecánicas y el comportamiento de autoensamblaje de estos hidrogeles pentapeptídicos.
    • Evaluar la citocompatibilidad y las capacidades protectoras de estos hidrogeles para la administración celular y la ingeniería de tejidos neurales.

    Principales métodos:

    • La reología y la espectroscopia se utilizaron para analizar las propiedades mecánicas y la influencia de las variaciones de secuencia, pH y concentración.
    • Microscopía electrónica de transmisión (TEM) y microscopía cryoelectrónica (Cryo-EM) visualizan la morfología de las fibrillas.
    • Las simulaciones de dinámica molecular corroboraron el comportamiento experimental de la gelación.
    • Se realizaron ensayos de encapsulación y proliferación celular utilizando células progenitoras de oligodendrocitos (OPC).

    Principales resultados:

    • Siete péptidos no modificados formaron hidrogeles robustos (0,2 - 20 kPa) en bajas concentraciones (< 3 por ciento en peso) con propiedades de adelgazamiento y rápida autocuración.
    • Los péptidos se autoensamblan en fibrillas retorcidas en forma de cinta (decenas de nanómetros de diámetro) con periodicidades similares a las amiloides.
    • Los hidrogeles apoyaron el encapsulamiento, la proliferación y la extensión del proceso 3D de OPC.
    • Los hidrogeles protegieron a los OPC de la tensión mecánica durante la inyección, demostrando su potencial como portadores de células.

    Conclusiones:

    • Los pentapéptidos de ensamblaje rápido para administración inyectable (RAPID) desarrollados presentan propiedades mecánicas y estructurales ajustables.
    • Estos hidrogeles son adecuados para la administración celular y la ingeniería de tejidos neuronales debido a su inyectabilidad, citocompatibilidad y capacidades protectoras.
    • El autoensamblaje de estos péptidos cortos en hidrogeles de nanofibra funcionales abre nuevas vías para la medicina regenerativa.
