  • Los GPCR muestran una mayor afinidad por los agonistas en su conformación activa en comparación con su estado inactivo.
  • Los mecanismos moleculares que subyacen a esta diferencia de afinidad siguen siendo incompletamente entendidos.

Objetivo del estudio:

  • Esclarecer la base estructural para el aumento de la afinidad agonista en el estado activo de los GPCR.
  • Comparar los estados activo e inactivo del receptor β1 (β1AR) unido a los agonistas.

Principales métodos:

  • Se determinaron cuatro estructuras de estado activo de β1AR complejas con nanobodies específicos de la conformación y agonistas de eficacia variable.
  • Se compararon estas estructuras de estado activo con estructuras de estado inactivo previamente determinadas de β1AR unidas a los mismos ligandos.

Principales resultados:

  • Se observó una reducción significativa (24-42%) en el volumen del sitio de unión ortostérico en el estado activo en comparación con el estado inactivo.
  • Se encontró que los enlaces de hidrógeno potenciales entre el receptor y el ligando eran más cortos en el estado activo.
  • Se observó un aumento (hasta un 30%) en los contactos atómicos entre el β1AR y el ligando en la conformación activa.

Conclusiones:

  • El bolsillo de unión más pequeño y más restringido en el estado activo de β1AR conduce a mayores contactos atómicos y enlaces de hidrógeno más cortos.
  • Estos cambios estructurales proporcionan una explicación molecular para la mayor afinidad agonista observada en los GPCR activos.
  • Los hallazgos son aplicables a una amplia gama de agonistas estructuralmente diversos que se unen a los GPCR.
