Sus antecedentes:

Objetivo del estudio:

  • Desarrollar una nueva estrategia para sondear las estructuras de glicano bacteriano que son difíciles de estudiar utilizando la incorporación metabólica.

  • Generación de fosfolípidos de arabinofuranosilo sintéticos como donantes de glicolipidos no naturales
  • Prueba de los donantes sintéticos en modelos de Corynebacterium glutamicum y Mycobacterium smegmatis.
  • Utilizando un C. glutamicum mutante que carece de arabinan para experimentos de complementación química.
  • El uso de sustratos de glicano etiquetados isotópicamente para la caracterización de la pared celular a través de RMN.
  • Evaluación del procesamiento de donantes sintéticos por las arabinofuranosil transferasas y su efecto en la recuperación de la envoltura celular.

Principales resultados:

  • Los glucolipidos sintéticos no naturales sirvieron con éxito como intermediarios biosintéticos, restaurando la arabinana de la pared celular en un mutante de C. glutamicum.
  • La adición de una sonda sintética basada en lípidos permitió una caracterización completa de la pared celular utilizando RMN.
  • Las cinco arabinofuranosil transferasas conocidas procesaron el azúcar donante exógeno ligado a lípidos, lo que condujo a la recuperación completa de la envoltura celular.
  • La sonda basada en lípidos podría rescatar células salvajes tratadas con un inhibidor de la biosíntesis de la pared celular.

Conclusiones:

  • Los sustitutos de los glicanos ligados a los lípidos naturales pueden intervenir eficazmente en las vías de biosíntesis de los glicanos celulares.
  • La incorporación biosintética de glicanos vinculados a lípidos sintéticos es una estrategia poderosa para sondear la estructura y la función de los glicanos en las bacterias.
  • Este enfoque ofrece una alternativa valiosa para el estudio de estructuras complejas de glicanos recalcitrantes a los métodos tradicionales.
