Sus antecedentes:

  • La iniciación de la traducción bacteriana implica la formación de un complejo 70S a partir de las subunidades 30S y 50S, el tRNA iniciador y los factores.
  • Los factores de iniciación deben disociarse para que el complejo 70S se alargue, pero la dinámica y los intermedios siguen sin estar claros.

Objetivo del estudio:

  • Elucidar los cambios conformacionales y los intermedios durante la formación y maduración del complejo de iniciación 70S.
  • Comprender el papel de la dinámica en la regulación de la iniciación de la traducción bacteriana.

Principales métodos:

  • Se empleó microscopía electrónica criogénica de resolución temporal (TR-cryo-EM) para capturar instantáneas de resolución casi atómica.
  • El análisis se centró en los reordenamientos conformacionales del ribosoma, los factores de iniciación y el fMet-tRNA.

Principales resultados:

  • Visualizó una serie ordenada en el tiempo de cambios conformacionales que impulsan la unión de subunidades y la disociación de factores.
  • Capturó las reorganizaciones dinámicas del posicionamiento de fMet-tRNA durante la maduración al estado competente para la elongación.

Conclusiones:

  • TR-cryo-EM proporciona una visión sin precedentes en el mecanismo y la regulación de la formación del complejo 70S.
  • El estudio aclara el proceso dinámico de iniciación de la traducción bacteriana y la competencia de alargamiento.
