Resumen

Sus antecedentes:

  • Comúnmente atribuido a la actividad antioxidante, este estudio investiga esta suposición.

Objetivo del estudio:

  • Investigar el mecanismo por el cual el resveratrol afecta la presión arterial.
  • Para determinar si el efecto reductor de la presión arterial del resveratrol está relacionado con la oxidación de las proteínas.
  • Para aclarar el papel de la PKG1α (proteína quinasa 1α) dependiente del cGMP en la acción del resveratrol.

Principales métodos:

  • Se evaluó la capacidad del resveratrol para oxidar el PKG1α dependiente del cGMP en las células del músculo liso aórtico de la rata.
  • Se utilizaron experimentos in vitro, espectrometría de masas y resonancia paramagnética de electrones para la caracterización mecánica.
  • Monitoreo de la presión arterial en ratones tipo salvaje y en ratones knock-in C42S durante el tratamiento con hipertensión inducida y resveratrol.

Principales resultados:

  • La oxidación del resveratrol de los anillos fenólicos paradójicamente causa la oxidación de las proteínas a través de una quinona reactiva.
  • Este proceso oxida los residuos de cisteína, un efecto amplificado bajo estrés oxidativo.
  • La activación oxidativa inducida por resveratrol de PKG1α redujo la presión arterial en ratones hipertensos, pero no en ratones con PKG1α resistente.

Conclusiones:

  • El resveratrol reduce la presión arterial a través de un mecanismo que involucra la oxidación paradójica de las proteínas.
  • Este efecto es particularmente pronunciado durante el estrés oxidativo.
  • Los hallazgos sugieren que la oxidación de proteínas puede ser un mecanismo común para moléculas con propiedades antioxidantes.
