Sus antecedentes:

  • Se conocen las funciones de los macrófagos en la lesión miocárdica, pero los neutrófilos en la patología cardíaca no isquémica son poco estudiados.
  • La investigación de la función de los neutrófilos en la hipertrofia cardíaca inducida por sobrecarga de presión es crucial para comprender la insuficiencia cardíaca.

Objetivo del estudio:

  • Para aclarar el papel y la regulación de los neutrófilos en la hipertrofia cardíaca inducida por sobrecarga de presión.
  • Para determinar la participación de la señalización de Wnt5a en la infiltración de neutrófilos y la patología cardíaca.

Principales métodos:

  • Los ratones fueron tratados con anticuerpos Ly6G para agotar los neutrófilos.
  • Los ratones se sometieron a una constricción aórtica transversal (TAC) para inducir una sobrecarga de presión.
  • La expresión de Wnt5a fue manipulada en las células mieloides (deficiencia o sobreexpresión).

Principales resultados:

  • La depleción de neutrófilos atenuó la hipertrofia cardíaca inducida por TAC, la inflamación y preservó la función cardíaca.
  • La deficiencia de Wnt5a mieloide suprimió la infiltración de neutrófilos y imitó los efectos neutropénicos.
  • La sobreexpresión de Wnt5a en las células mieloides exacerbó la hipertrofia cardíaca, la inflamación y la disfunción.

Conclusiones:

  • La infiltración de neutrófilos regulada por Wnt5a es crítica en la insuficiencia cardíaca inducida por sobrecarga de presión.
  • La orientación de la infiltración de neutrófilos puede ofrecer estrategias terapéuticas para la hipertrofia cardíaca y la insuficiencia cardíaca.
