Sus antecedentes:

  • La 5-formilcitosina (5fC) es una nucleobasa epigenética crucial en los genomas de los mamíferos, que actúa como un intermediario en la desmetilación activa del ADN.
  • Las interacciones únicas de 5fC con las proteínas nucleares sugieren funciones significativas en la regulación del genoma.
  • Los métodos actuales para detectar 5fC enfrentan desafíos debido a su baja abundancia genómica y la necesidad de sondas selectivas.

Objetivo del estudio:

  • Desarrollar repeticiones TALE capaces de reconocimiento selectivo de 5fC y de enlace cruzado covalente.
  • Para superar las limitaciones de sensibilidad en la detección de niveles bajos de 5fC en el ADN genómico.
  • Para permitir la cuantificación directa de los niveles de 5fC utilizando sondas programables.

Principales métodos:

  • Efectores similares al activador de transcripción (TALEs) diseñados con p-acetilfenilalanina (pAcF) para la unión universal a la citosina.
  • Utilizó el enlace cruzado de dioximas mediado por diaminooxy-linker para la formación selectiva de aductos de 5fC.
  • Combinaciones de enlaces repetidos optimizadas para la resolución de un solo sitio CpG.

Principales resultados:

  • Se ha demostrado la unión universal de TALEs diseñados a todas las nucleobases de citosina de mamíferos.
  • Se ha logrado el enlace cruzado covalente selectivo específicamente a 5fC.
  • Se cuantificaron con éxito los niveles de 5fC en un fondo del genoma humano mediante enriquecimiento covalente.
  • Se han identificado combinaciones específicas de repetición y enlace TALE para la detección de 5fC de alta resolución.

Conclusiones:

  • Desarrolló una nueva estrategia basada en TALE para el enlace covalente y la detección de 5fC.
  • Este método mejora la sensibilidad y la selectividad para analizar las modificaciones epigenéticas.
  • Amplia la utilidad de las sondas programables para estudios epigenéticos más allá de las nucleobases estándar (A, G, T, C).
