Sus antecedentes:

  • El papel de la expresión génica en el desarrollo de los órganos de los mamíferos es poco conocido.
  • Faltan datos transcriptómicos comparativos entre especies y etapas de desarrollo.

Objetivo del estudio:

  • Caracterizar la evolución de la expresión génica durante el desarrollo de órganos de mamíferos.
  • Proporcionar un recurso completo de transcriptomas del desarrollo para el análisis comparativo.

Principales métodos:

  • Se recogieron transcriptomas de siete órganos (cerebro, cerebelo, corazón, riñón, hígado, ovario y testículo).
  • Análisis de datos a través de múltiples puntos de tiempo de desarrollo y siete especies (humano, macaco rhesus, ratón, rata, conejo, opossum, pollo).
  • Modelos de expresión génica comparados para identificar las correspondencias de las etapas de desarrollo y las diferencias evolutivas.

Principales resultados:

  • Identificaron etapas de desarrollo conservadas en todas las especies y tiempos distintos en el desarrollo de las gónadas.
  • Se observó una disminución en la amplitud de la expresión génica y la selección purificadora, con un aumento en la selección positiva y la nueva expresión génica durante el desarrollo.
  • Se encontraron variaciones significativas en las trayectorias temporales de expresión génica entre especies, con tejidos cerebrales que muestran estabilidad y hígado / testículos que muestran cambios dinámicos.

Conclusiones:

  • Este estudio proporciona un recurso valioso para comprender el desarrollo y la evolución de los órganos de los mamíferos.
  • La transcriptómica comparativa revela cambios dinámicos en las presiones de selección y la expresión génica durante el desarrollo.
  • Las diferencias en el tiempo de desarrollo y las trayectorias de expresión génica resaltan los caminos evolutivos específicos de las especies.
