Sus antecedentes:

Objetivo del estudio:

  • Investigar los cambios relacionados con la edad en los nichos neurogénicos de ratones utilizando la secuenciación de ARN de una sola célula.
  • Identificar las interacciones celulares y moleculares que contribuyen al deterioro neurogénico relacionado con la edad.

Principales métodos:

  • Secuenciación de ARN unicelular de nichos neurogénicos de ratones jóvenes y viejos.
  • Análisis de 14.685 transcriptomas de una sola célula.
  • Experimentos de cocultivo in vitro e in vivo para evaluar las interacciones de las células T y las células madre neurales.

Principales resultados:

  • Los nichos neurogénicos viejos mostraron reducción de células madre neurales activadas, células endoteliales y microglia alteradas, y infiltración de células T.
  • Las células T que se infiltran en el cerebro se expanden clonalmente, son distintas de las células T de la sangre y expresan interferón-γ.
  • Las células madre neurales sensibles al interferón-γ presentaron una disminución de la proliferación.
  • Se encontró que las células T inhiben la proliferación de células madre neurales a través de la secreción de interferón-γ.

Conclusiones:

  • Se identificó una nueva interacción entre las células T y las células madre neurales en los cerebros envejecidos.
  • La inhibición mediada por las células T, en parte a través del interferón-γ, contribuye a la disminución de la proliferación de células madre neurales relacionada con la edad.
  • Estos hallazgos sugieren posibles objetivos terapéuticos para contrarrestar la disfunción cerebral relacionada con la edad.
