Reporter Genes

Reporter genes are a type of protein-coding gene that are often tagged to a gene of interest. Once inside a target cell, reporter genes usually produce visually identifiable characteristics like fluorescence and luminescence when expressed along with the gene of interest. Thus, reporter genes “report” the presence or absence of genes of interest in an organism, determine the gene expression pattern, or track the physical location of a DNA segment or protein in the cell.

Resumen

El proto-oncogén c-fos, cuando se sobreexpresa en ratones transgénicos, conduce a agrandamiento del bazo y del timo, afectando el desarrollo de las células T y causando inmunodeficiencia. Esto sugiere que el c-fos afecta indirectamente la maduración de las células T al estimular las células epiteliales del timo.

Sus antecedentes:

  • Comprender la función de c-fos en los tejidos hematopoyéticos es crucial para comprender el desarrollo del sistema inmunológico y la potencial oncogénesis.

Objetivo del estudio:

  • Investigar el papel del protooncogén c-fos en los tejidos hematopoyéticos utilizando modelos de ratón transgénicos.
  • Para determinar los efectos de la expresión exógena de c-fos en el desarrollo de las células T y la función inmune.

Principales métodos:

  • Generación de ratones transgénicos (H2-c-fos) que expresan c-fos bajo el promotor H2-Kb.
  • Análisis de la morfología del bazo y del timo, las poblaciones celulares y los linajes hematopoyéticos.
  • Utilizando quimeras de radiación de médula ósea para evaluar los efectos específicos de las células.

Principales resultados:

  • Los ratones H2-c-fos exhibieron agrandamiento del bazo y del timo hiperplásico con aumento de las células epiteliales del timo.
  • La expresión exógena de c-fos alteró el desarrollo de las células T, aumentando los timocitos maduros pero deteriorando la función de las células B y T, lo que condujo a la inmunodeficiencia.
  • La distribución alterada del subconjunto de células T no fue un efecto directo de c-fos en las células T, lo que sugiere mecanismos indirectos.

Conclusiones:

  • La sobreexpresión de c-fos en ratones transgénicos conduce a la proliferación de células epiteliales del timo, lo que afecta indirectamente el desarrollo de las células T.
  • El estudio revela un papel complejo para c-fos en la regulación inmune, destacando su potencial para inducir inmunodeficiencia en lugar de malignidad en este contexto.
