Sus antecedentes:

Objetivo del estudio:

  • Investigar el mecanismo de acción de los anticuerpos alfa-4-beta-7 de la integrina en el control de la infección por SIV.
  • Comparar los efectos de un anticuerpo alfa-4-beta-7 integrina con los efectos pleiotrópicos frente a los anticuerpos monoclonales que sólo bloquean la unión a SIV Env.

Principales métodos:

  • Treinta macacos rhesus fueron infectados con una cepa SIV atenuada (nef-atenuada).
  • Todos los macacos recibieron terapia antirretroviral.
  • Los animales se dividieron en cinco grupos, cada uno de los cuales recibió diferentes tratamientos con anticuerpos, incluyendo un anticuerpo anti-alfa-4-beta-7 integrina y anticuerpos no neutralizantes dirigidos a SIV Env.

Principales resultados:

  • A diferencia de un informe anterior, este estudio no encontró control virológico sostenido en ninguno de los grupos de tratamiento.
  • La comparación entre el anticuerpo pleiotrópico y los anticuerpos bloqueantes no reveló una diferencia en el control virológico.
  • Estos hallazgos sugieren que los tratamientos con anticuerpos probados, en combinación con ART, fueron ineficaces para lograr un control duradero del SIV en este modelo.

Conclusiones:

  • La combinación de terapia antirretroviral y anticuerpos dirigidos a la alfa-4-beta-7 o anticuerpos bloqueadores no condujo a un control sostenido del SIV en macacos rhesus.
  • El mecanismo de acción sugerido por estudios anteriores no se replicó, lo que pone de relieve la complejidad de la patogénesis y el tratamiento del SIV/ VIH.
