Sus antecedentes:

  • Las interacciones receptor-ligando (IRL) son cruciales en biología, pero complejas en las superficies celulares debido a la distribución dinámica de los receptores.
  • El modelo tradicional de llaves y cerraduras describe inadecuadamente estas RLI complejas de la superficie celular.

Objetivo del estudio:

  • Desarrollar una plataforma programada de marco de ADN tetraédrico (TDF) para RLI de superficie celular de ingeniería topológica.
  • Para permitir la captura de alta afinidad de las células tumorales circulantes (CTC) mediante el reclutamiento activo de receptores agrupados.

Principales métodos:

  • Síntesis de n-simplexos mediante TDF con 1 a 3 aptameros dirigidos a la molécula de adhesión de las células epiteliales (EpCAM).
  • Utilizó el anclaje ortogonal de ligandos en vértices TDF para la organización espacial.
  • Se utilizaron sondas 2-simplex para la captura de CTC y se evaluó su afinidad de unión y su efecto sobre la endocitosis.

Principales resultados:

  • El 2-simplex con tres aptameros EpCAM demostró un aumento de ~19 veces en la afinidad de unión.
  • Los RLI diseñados impidieron la endocitosis de las células cancerosas.
  • Se logró una captura de CTC de alta eficiencia a partir de muestras de pacientes con cáncer de mama clínico.

Conclusiones:

  • La plataforma programada con TDF ofrece un método poderoso para la ingeniería de RLI de superficie celular.
  • Este enfoque facilita el estudio de las RLI en contextos fisiológicos.
  • La plataforma muestra un potencial significativo para avanzar en el diagnóstico del cáncer, en particular para la detección de CTC.
