Sus antecedentes:

  • Los ácidos biliares son abundantes en el intestino de los mamíferos y son transformados por las bacterias en moléculas bioactivas.
  • Si bien se sabe que influyen en el metabolismo, el cáncer y la inmunidad innata, su impacto en las células inmunes adaptativas como las células T auxiliares 17 (T H 17) y las células T reguladoras (T reg) sigue sin estar claro.

Objetivo del estudio:

  • Investigar los efectos de los metabolitos del ácido biliar en las células inmunes adaptativas, específicamente las células TH17 y Treg.
  • Identificar derivados específicos de ácido biliar que modulan la diferenciación y la función de las células T.

Principales métodos:

  • Selección de una biblioteca de metabolitos de ácido biliar.
  • Evaluación del impacto de los metabolitos identificados en la diferenciación celular TH17 y Treg in vitro e in vivo.
  • Investigar los mecanismos moleculares de acción, incluida la unión al factor de transcripción y la producción de especies reactivas de oxígeno.

Principales resultados:

  • Se identificaron dos derivados del ácido litólico (LCA), 3-oxoLCA e isoalloLCA, como reguladores de las células T.
  • 3-OxoLCA inhibió la diferenciación de las células TH17 al unirse a RORγt.
  • IsoalloLCA promovió la diferenciación de células Treg a través de la producción mitocondrial de ROS y requirió el potenciador del SNC3 para su acción.

Conclusiones:

  • Los metabolitos del ácido biliar modulan directamente el equilibrio entre las células TH17 y las células Treg, influyendo en las respuestas inmunes del huésped.
  • Los derivados específicos del ácido biliar ofrecen mecanismos potenciales para controlar la inmunidad adaptativa a través de la modulación dirigida de las células T.
