Sus antecedentes:

  • La tiroglobulina (TG) es crucial para la síntesis de la hormona tiroidea, la regulación del metabolismo, el crecimiento y el desarrollo.
  • La producción de hormonas implica la iodación de tirosina y el acoplamiento dentro de la TG, pero los sitios específicos y los mecanismos estructurales siguen sin estar claros.
  • La falta de una estructura 3D TG ha obstaculizado la comprensión mecánica de la biosíntesis de la hormona tiroidea.

Objetivo del estudio:

  • Determinar la estructura tridimensional de alta resolución de la tiroglobulina humana de cuerpo entero.
  • Identificar y caracterizar los pares específicos de tirosina responsables de la síntesis de las hormonas tiroideas.
  • Aclarar las características estructurales y dinámicas que rigen la actividad de la tirosina hormonogénica.

Principales métodos:

  • Se utilizó microscopía criolectrónica para determinar la estructura de 3,5 Å de la tiroglobulina humana.
  • Se utilizaron ensayos de mutagénesis dirigida al sitio y de producción de hormonas in vitro para verificar los pares de tirosina identificados.
  • Se realizó la transferencia funcional de los sitios de reacción identificados a una proteína modificada (proteína de unión a la maltosa).

Principales resultados:

  • Se resolvió con éxito la estructura tridimensional de la tiroglobulina humana de longitud completa.
  • Se identificaron todos los pares de tirosina hormonogénica dentro de la estructura TG.
  • Las características clave de los sitios hormonogénicos incluyen la proximidad, la flexibilidad y la exposición al disolvente.
  • Los pares de donantes y receptores de tirosina diseñados en la proteína de unión a la maltosa imitaron la eficiencia de la producción de hormonas de TG.

Conclusiones:

  • El estudio proporciona la primera estructura 3D de la tiroglobulina humana, revelando detalles críticos de la síntesis de la hormona tiroidea.
  • La proximidad, la flexibilidad y la exposición al disolvente se identifican como determinantes clave de los sitios de tirosina hormonogénica.
  • Los hallazgos ofrecen un nuevo marco para comprender la producción y regulación de la hormona tiroidea, con posibles implicaciones terapéuticas.
