Antibodies, also known as immunoglobulins, are produced by B cells in response to foreign substances, such as bacteria and viruses. These proteins are critical for recognizing and neutralizing these substances, protecting the body from potential harm.
The basic structure of an antibody consists of four protein chains: two identical heavy chains and two identical light chains. These chains are held together by disulfide bonds and other non-covalent interactions, forming a Y-shaped structure.

  • La inmunoglobulina M (IgM) es crucial para la inmunidad humoral y mucosa.
  • El ensamblaje y el transporte de IgM involucran la cadena de unión (cadena J) y el receptor polimérico de inmunoglobulina (pIgR).
  • Los mecanismos moleculares subyacentes al ensamblaje y transporte de IgM siguen siendo poco conocidos.

Objetivo del estudio:

  • Para aclarar la base estructural del montaje y el transporte de IgM.
  • Para determinar las interacciones moleculares entre IgM-Fc, la cadena J y el ectodominio pIgR.

Principales métodos:

  • Para visualizar el complejo se empleó la crio-microscopía electrónica (crio-EM).
  • Análisis estructural de la región humana IgM-Fc en complejo con la cadena J y el ectodominio pIgR.

Principales resultados:

  • La estructura cryo-EM reveló un pentámero IgM-Fc asimétrico con una formación triangular hexagonal e incompleta.
  • Las piezas traseras IgM-Fc forman una estructura tipo amiloide para la estabilización del pentámero.
  • La cadena J une IgM-Fc y pIgR, induciendo un cambio conformacional significativo en pIgR para el compromiso complejo.

Conclusiones:

  • El estudio proporciona los primeros conocimientos estructurales sobre el complejo IgM-Fc, la cadena J y la pIgR.
  • Esta comprensión estructural ofrece una base para descifrar los mecanismos funcionales de la IgM en las respuestas inmunes.
