Sus antecedentes:

  • El colesterol es vital para las membranas celulares, pero está estrechamente regulado en el retículo endoplasmático (RE).
  • La enzima ER esterol O-aciltransferasa 1 (ACAT1) controla los niveles de colesterol mediante la formación de ésteres colesterol.
  • La desregulación de ACAT1 está relacionada con el Alzheimer, la aterosclerosis y los cánceres.

Objetivo del estudio:

  • Para aclarar la base estructural de la función y la inhibición de ACAT1.
  • Para comprender la interacción entre ACAT1 y su inhibidor, nevanimiba.
  • Proporcionar ideas para el desarrollo de nuevas terapias dirigidas a ACAT1.

Principales métodos:

  • Se utilizó la criomicroscopia electrónica (Cryo-EM) para determinar la estructura del ACAT1 humano.
  • Se realizaron análisis bioquímicos para estudiar la actividad enzimática y la unión al inhibidor.
  • Los datos estructurales se integraron con el conocimiento existente de los mecanismos catalíticos de ACAT1.
