Resumen

Objetivo del estudio:

  • Realizar un análisis exhaustivo de la variación estructural en un conjunto diverso de datos de la población humana.
  • Identificar nuevas variantes estructurales y comprender su distribución y frecuencia en las poblaciones globales.
  • Evaluar las limitaciones del genoma humano de referencia actual y la necesidad de datos genómicos diversos.

Principales métodos:

  • Análisis de los datos de secuenciación de alta cobertura del panel de Diversidad del Genoma Humano (911 muestras, 54 poblaciones).
  • Identificación y caracterización de las variantes estructurales, incluidas las duplicaciones e inserciones.
  • Ensamblaje de novo de 25 genomas utilizando secuenciación de lectura vinculada para descubrir inserciones resueltas en el punto de ruptura.

Principales resultados:

  • Se identificaron 126.018 variantes estructurales, de las cuales el 78% son nuevas en comparación con los proyectos globales anteriores.
  • Descubrió variantes específicas de la población, incluyendo duplicaciones fuera de control y segmentos introgresivos de homínidos arcaicos.
  • Se caracterizaron 1.643 inserciones únicas (1.9 Mb de secuencia total) ausentes del genoma de referencia GRCh38.

Conclusiones:

  • El estudio subraya la inmensidad de la variación estructural no descubierta en el genoma humano.
  • Las variantes específicas de la población revelan historias y adaptaciones evolutivas únicas.
  • Un solo genoma de referencia es insuficiente; los genomas diversos y de alta calidad son cruciales para una comprensión completa de la variación genética humana.
