Sus antecedentes:

  • Las proteínas que se unen a secuencias específicas de ADN utilizan difusión 3D y deslizamiento 1D en ADN no específico.
  • Comprender la dinámica de este deslizamiento en 1D es crucial para comprender la exploración del genoma.

Objetivo del estudio:

  • Caracterizar la dinámica de exploración del ADN de las moléculas individuales del lacrepresor (LacI) durante el deslizamiento en 1D.
  • Para investigar el movimiento de rotación y el comportamiento de salto de ranura de LacI en el ADN.

Principales métodos:

  • Se utilizó el seguimiento láser confocal de una sola molécula en tiempo real combinado con la espectroscopia de correlación de fluorescencia (SMCT-FCS) para monitorear la rotación de LacI en la escala de tiempo de microsegundos.
  • Se utiliza la transferencia de energía de resonancia por fluorescencia de una sola molécula (smFRET) para observar directamente los eventos de derivación de secuencias.

Principales resultados:

  • LacI exhibe deslizamiento acoplado por rotación, atravesando aproximadamente 40 pares de bases (bp) por revolución, excediendo el tono helicoidal del ADN.
  • Se ha observado que LacI salta de una a dos ranuras (10-20 pb) cada 200-700 μs, lo que indica un desvío frecuente de los sitios objetivo.
  • Se demostró una compensación entre la velocidad de deslizamiento y la precisión de búsqueda debido a las débiles interacciones proteína-ADN no específicas.

Conclusiones:

  • El mecanismo de deslizamiento 1D de LacI implica frecuentes saltos de ranura, equilibrando la velocidad de búsqueda con el posible desvío del objetivo.
  • La técnica SMCT-FCS desarrollada ofrece una herramienta poderosa para investigar la dinámica molecular en la escala de tiempo de microsegundos en las interacciones biológicas.
