Sus antecedentes:

  • Las interacciones físicas entre los elementos reguladores distales son vitales para la regulación de la expresión génica.
  • La especificidad del tipo celular y la contribución de estas interacciones a la expresión génica siguen siendo incompletamente comprendidas.

Objetivo del estudio:

  • Mapear los bucles de cromatina mediados por cohesión y analizar la expresión génica en diversos tipos de células humanas.
  • Investigar la variación de tipo celular en los bucles de cromatina y su correlación con los patrones de expresión génica.

Principales métodos:

  • Se utilizó el análisis de interacción de la cromatina por secuenciación de etiquetas de extremo emparejado (ChIA-PET) para mapear los bucles de cromatina mediados por la cohesión.
  • Se analizaron datos de expresión génica de 24 tipos de células humanas diferentes como parte del proyecto ENCODE.

Principales resultados:

  • Se identificó que el 28% de todos los bucles de cromatina exhiben variación en diferentes tipos de células.
  • Se observó una correlación modesta entre las variaciones del bucle de cromatina y los cambios en la expresión génica.
  • Demostró que los patrones de bucle de cromatina agrupan eficazmente los tipos de células en función de su tejido de origen.

Conclusiones:

  • Los bucles de cromatina específicos del tipo de célula juegan un papel en la regulación de la expresión génica.
  • Los patrones de conectividad genética se correlacionan con las clases de genes funcionales, como los genes sensibles a la dosis.
  • Este atlas de bucles de cromatina mejora la comprensión de la estructura y función del genoma dentro de la Enciclopedia ENCODE.
