  • La resonancia magnética nuclear bidimensional (2D NMR) es estándar para estudiar los procesos de intercambio (cambio de conformación, protonación, unión) en escalas de tiempo de milisegundos a través del análisis de picos cruzados.
  • Las técnicas de resonancia de espín de electrones (ESR), como 2D-ELDOR, ofrecen potencial para estudiar procesos de intercambio más rápidos (de nanosegundos a microsegundos), pero están limitadas por la resolución de la señal para sondas comunes como los nitróxidos.
  • El aumento de la resolución del factor g en frecuencias ESR más altas (por ejemplo, 95 GHz) puede superar las limitaciones en la resolución de estados de intercambio dentro de sus anchos de línea.

Objetivo del estudio:

  • Demostrar la capacidad de 95 GHz 2D-ELDOR para el estudio de los procesos de intercambio químico que implican etiquetas de giro de óxido de nitrógeno.
  • Analizar el desarrollo transversal derivado del intercambio químico en sistemas relevantes para los estudios biofísicos y químicos.

Principales métodos:

  • Se empleó la doble resonancia electrón-electrón bidimensional (2D-ELDOR) para observar el desarrollo de picos cruzados.
  • Estudió una etiqueta de imidazolina sensible al pH y una partición de radicales de nitrógeno entre las fases acuosa y lipídica.

Principales resultados:

  • Se ha observado y analizado con éxito el desarrollo de picos cruzados debido al intercambio químico en etiquetas de espín de óxido de nitrógeno a 95 GHz.
  • Se ha demostrado el estudio de la dinámica de protonación/deprotonación de una etiqueta de espín de imidazolina, controlada por el pH y la concentración del tampón.
  • Se ha presentado el análisis del intercambio de radicales de óxido de nitrógeno entre los ambientes acuoso y fosfolípido en las vesículas lipídicas.

Conclusiones:

  • 95 GHz 2D-ELDOR es una poderosa técnica para la investigación en tiempo real de los procesos de intercambio químico de nanosegundos a microsegundos que involucran etiquetas de espín de óxido de nitrógeno.
  • Este trabajo establece la primera observación y análisis de picos cruzados de intercambio químico utilizando etiquetas de espín de óxido de nitrógeno en ESR.
  • La metodología desarrollada abre nuevas vías para el estudio de la dinámica molecular en diversos sistemas químicos y biológicos.
