Sus antecedentes:

  • La endonucleasa RAG inicia la recombinación Igh V(D) J mediante el escaneo de la cromatina en busca de secuencias de señales de recombinación (RSS).
  • La extrusión de bucle mediada por cohesión facilita el escaneo RAG de largo alcance, mientras que los elementos de unión CTCF (CBEs) pueden obstaculizar este proceso.
  • El locus V(H) sufre una contracción poco comprendida en las células B progenitoras, lo que potencialmente mejora la accesibilidad distal V(H).

Objetivo del estudio:

  • Aclarar los mecanismos subyacentes a la contracción del locus V ((H) y su impacto en el escaneo RAG.
  • Investigar el papel de la extrusión de bucle y las CBE en la regulación de la recombinación V ((D) J en las células B progenitoras.

Principales métodos:

  • Análisis de una inversión del locus V (H) en células B progenitoras primarias de ratón.
  • Evaluación de V(H) - RSS y el reordenamiento críptico de RSS.

Principales resultados:

  • La inversión de locus V ((H) abrogó V ((H) -RSS y el reordenamiento críptico normalmente convergente de RSS, a pesar de la contracción de locus.
  • Inversión activada por la utilización de criptografía orientada en sentido opuesto V{\displaystyle \mathrm {V} }-RSS y RAG escaneando más allá del locus V{\displaystyle \mathrm {R} } .
  • La baja expresión de WAPL en las células B progenitoras primarias se correlacionó con la contracción del locus y el escaneo RAG; el agotamiento de WAPL activó estos procesos.

Conclusiones:

  • La desregulación de los impedimentos CBE promueve el escaneo RAG del locus V ((H) a través de la extrusión en bucle en las células B progenitoras.
  • La extrusión de bucle está implicada en la contracción del locus V (H), con WAPL influyendo en la dinámica de la cohesión.
  • Estos hallazgos proporcionan una visión mecanicista de la accesibilidad del locus V (H) y la regulación de la recombinación V (D) J.
