Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.

Resumen

Sus antecedentes:

  • TP53 (proteína tumoral p53) es el gen conductor del cáncer más frecuentemente mutado.
  • Dirigirse a los genes supresores de tumores mutados, incluido el TP53, sigue siendo un desafío significativo en la terapia del cáncer debido a la falta de medicamentos disponibles.

Objetivo del estudio:

  • Identificar un anticuerpo altamente específico para la mutación común TP53 R175H presente en la superficie celular.
  • Desarrollar un agente inmunoterapéutico basado en este anticuerpo para atacar cánceres difíciles de tratar.

Principales métodos:

  • Identificación de un anticuerpo con alta especificidad para la mutación TP53 R175H en complejo con el antígeno leucocitario humano-A (HLA-A).
  • Elucidación de la base estructural para la especificidad de los anticuerpos.
  • La ingeniería del anticuerpo en un diacuerpo bispecífico de cadena única para aplicación terapéutica.
  • Pruebas in vitro e in vivo (en ratones) de la eficacia del anticuerpo bispecífico en la activación de las células T y la lisía de las células cancerosas.

Principales resultados:

  • Se identificó un anticuerpo que reconoce específicamente el neoantígeno TP53 R175H presentado por HLA-A en las células cancerosas.
  • El diabody bispecífico de cadena única activó eficazmente las células T para lisar las células cancerosas que presentan el neoantígeno objetivo, incluso a bajas densidades complejas.
  • Eficacia demostrada in vitro e in vivo del enfoque inmunoterapéutico.

Conclusiones:

  • Se ha desarrollado un nuevo anticuerpo bispecífico dirigido a la mutación TP53 R175H común.
  • Este enfoque es prometedor para el tratamiento de cánceres con mutaciones que son convencionalmente difíciles de detectar.
  • La estrategia ofrece una nueva vía potencial para la inmunoterapia del cáncer.
