This lesson illustrates the role of deuterium substitution in simplifying the NMR spectrum of compounds comprising labile protons. One method employed is the use of deuterium. Amongst the three isotopes of hydrogen, deuterium (2H) has a nucleus composed of one proton and one neutron. Sus antecedentes:

  • Los reactivos electrófilos se utilizan comúnmente para el etiquetado de histidina.
  • Existe la necesidad de estrategias alternativas de funcionalización de la histidina, en particular las que permiten un etiquetado rápido y selectivo.

Objetivo del estudio:

  • Informar sobre una estrategia para la funcionalización de la histidina.
  • Demostrar el etiquetado rápido y selectivo de proteínas utilizando oxígeno (1O2).
  • Aplicar este método para el etiquetado de proximidad a escala nanométrica.

Principales métodos:

  • Se utilizó una pequeña molécula nucleofílica, 1-metil-4-arilurazol, para el etiquetado de histidina.
  • Oxígeno singlete generado (1O2) en condiciones controladas.
  • Se empleó un fotocatalizador localizado cerca de un sitio de unión de ligando para el etiquetado selectivo de anticuerpos Fc en cuentas magnéticas.

Principales resultados:

  • Logrado el etiquetado selectivo de histidina con el urazol nucleófilo bajo la generación de 1O2.
  • Se ha demostrado el etiquetado selectivo rápido de anticuerpos Fc utilizando un fotocatalizador localizado.
  • Se identificaron tres residuos específicos de histidina alrededor del sitio de unión mediante cromatografía líquida-espectrometría de masas.

Conclusiones:

  • La estrategia de umpolung desarrollada ofrece un nuevo enfoque para la funcionalización de la histidina.
  • El etiquetado de histidina mediada por oxígeno es efectivo para el etiquetado rápido y selectivo de proteínas.
  • Esta técnica es adecuada para aplicaciones de etiquetado de proximidad a escala nanométrica.
