Spontaneous mutations arise infrequently during DNA replication due to errors in the process. A key factor behind these errors is tautomeric shifts in nitrogenous bases, where bases transition from keto to enol forms or amino to imino forms. This shift can alter base-pairing rules, leading to mutations. Additionally, reactive oxygen species (ROS) arising from aerobic metabolism can damage DNA, resulting in depurination (loss of a purine base) or depyrimidination (loss of a pyrimidine base).

  • Para reconstruir las filogenias celulares tempranas en el desarrollo humano.
  • Investigar las tasas de mutación endógena y la dinámica clonal.
  • Para explorar las contribuciones celulares a diferentes tejidos y órganos.

Principales métodos:

  • Secuenciación de todo el genoma de colonias de una sola célula.
  • Secuenciación profunda de tejidos masivos de donantes humanos.
  • Utilizando mutaciones somáticas como códigos de barras para la reconstrucción filogenética.

Principales resultados:

  • Identificó una alta tasa de mutación inicial disminuyendo con el tiempo.
  • Contribución desigual demostrada de las primeras células al embrión propiamente dicho.
  • Se observaron desequilibrios clonales en diferentes lados del cuerpo, capas germinales y órganos.
  • Encontré evidencia de unas pocas células ancestrales que contribuyen significativamente a la sangre y al hígado de los adultos.
