Sus antecedentes:

  • La estructura tridimensional (3D) de la cromatina es crucial para la regulación genética y la función celular.
  • Los métodos existentes de captura de la conformación de la cromatina tienen limitaciones en el mapeo de los sistemas neuronales, particularmente con respecto a los cambios dinámicos y la especificidad del tipo celular sin disociación tisular.

Objetivo del estudio:

  • Desarrollar y aplicar un nuevo método, immunoGAM, para mapear la topología de la cromatina en 3D en todo el genoma en tipos específicos de células cerebrales de animales individuales sin alteración de los tejidos.
  • Investigar la relación entre las estructuras de cromatina 3D especializadas del tipo celular y los patrones de expresión génica en el cerebro.

Principales métodos:

  • ImmunoGAM, una extensión del mapeo de la arquitectura del genoma (GAM), utiliza tecnología libre de ligadura en criosecciones nucleares para mapear la topología del genoma.
  • Permite la selección del tipo celular a partir de tejidos complejos utilizando un número bajo de células (aprox. 1,000 células) y evita la disociación de los tejidos.
  • Las interacciones de cromatina se identifican mediante el análisis de la probabilidad de co-segregación de los loci de ADN a través de las rebanadas nucleares.

Principales resultados:

  • Las estructuras de cromatina 3D especializadas de tipo celular se mapearon en múltiples escalas genómicas, correlacionadas con los patrones de expresión génica.
  • Se observó una "fusión" extensa de genes largos durante la alta expresión y/o la alta accesibilidad a la cromatina.
  • Los contactos específicos del subtipo de neuronas involucran genes relacionados con la adicción y la plasticidad sináptica, con sitios de unión al factor de transcripción en la cromatina accesible.
  • Se encontraron genes de receptores sensoriales en compartimentos heterocromáticos, formando contactos de largo alcance.

Conclusiones:

  • ImmunoGAM proporciona una poderosa herramienta para estudiar la organización tridimensional de la cromatina en tipos específicos de células cerebrales a resolución de un solo animal.
  • Las conformaciones específicas de cromatina en las células cerebrales están intrincadamente relacionadas con los mecanismos de regulación genética y las funciones celulares especializadas.
  • Los hallazgos destacan la importancia de la arquitectura del genoma 3D en la identidad y función de las células neuronales.
