The theory of catalytically perfect enzymes was first proposed by W.J. Albery and J. R. Knowles in 1976. These enzymes catalyze biochemical reactions at high-speed. Their catalytic efficiency values range from 108-109 M-1s-1. These enzymes are also called 'diffusion-controlled' as the only rate-limiting step in the catalysis is that of the substrate diffusion into the active site. Objetivo del estudio:

  • Introducir y demostrar la catálisis sinérgica basado en ADN.
  • Combinar las capacidades organizativas del ADN con la catálisis sinérgica para mejorar el rendimiento.

Principales métodos:

  • Utilizó el ADN como un andamio para posicionar el cobre y los cocatalizadores TEMPO.
  • Se ha investigado la oxidación del alcohol aeróbico catalizado por Cu-TEMPO.
  • Se han incorporado horquillas de ADN para controlar la proximidad del cocatalizador.

Principales resultados:

  • Se obtiene un alto número de rotación del catalizador tras la dilución.
  • Demostró una mejora de 190 veces en el número de rotación del catalizador en comparación con los catalizadores sin andamio.
  • Control de las velocidades de reacción a través de cambios conformacionales en el ADN.

Conclusiones:

  • El andamiaje de ADN es compatible con la catálisis sinérgica.
  • Este enfoque mejora la eficiencia del catalizador y permite un control sensible al estímulo.
  • Abre nuevas posibilidades para el descubrimiento de la reacción, la detección y los materiales sensibles.
