Resumen
Los investigadores han desarrollado "drugtamers", moléculas similares al ADN hechas enteramente de fármacos contra el cáncer, que atacan y matan eficazmente a las células cancerosas. Este nuevo enfoque mejora significativamente la carga de fármacos y mejora la terapia dirigida contra el cáncer.

Sus antecedentes:
- Los conjugados de fármacos aptameros (ApDC) son prometedores para la terapia dirigida contra el cáncer debido a la unión específica de los aptameros.
- Los ApDC convencionales tienen limitaciones en la capacidad de carga de fármacos, ya que los aptámeros actúan principalmente como grupos objetivo con conjugación de fármacos limitada.
Objetivo del estudio:

- Crear oligómeros similares al ADN, llamados fármaco-támeros, mediante la sustitución de nucleósidos naturales por análogos de nucleósidos aprobados clínicamente.
Principales métodos:
- Se utilizaron cuatro análogos de nucleósidos aprobados clínicamente (clofarabina, ara-guanosina, gemcitabina, floxuridina) para sintetizar oligómeros similares al ADN constituidos por fármacos enteros (tameros de fármacos).
- Se evaluó la capacidad de orientación y la afinidad de unión de los fármaco-tameros a los receptores de la superficie de las células cancerosas.
- Se evaluó la eficacia de los fármaco-támeros en la inducción de la apoptosis de las células cancerosas y la inhibición de la progresión tumoral.
Principales resultados:
- Los aptameros del fármaco mantuvieron la capacidad de orientación de los aptameros progenitor, específicamente se unen a los receptores sobreexpresados en las células cancerosas.
- Los nuevos fármacos tamers lograron una relación de carga de fármaco del 100%.
- Se demostró una potente inducción de la apoptosis de las células cancerosas e inhibición de la progresión tumoral a través de la liberación enzimática del fármaco.
Conclusiones:
- Los drugtamers representan un avance significativo con respecto a los ApDC convencionales al ofrecer una alta carga de fármacos y una actividad terapéutica inherente.
- Esta estrategia de administración de fármacos tiene el potencial de mejorar la terapia dirigida al cáncer al combinar la orientación y la administración de fármacos potentes en una sola molécula.
