The pentose sugar in DNA is deoxyribose, while in RNA the pentose sugar is ribose. The difference between the sugars is the presence of the hydroxyl group on the ribose's second carbon and a hydrogen on the deoxyribose's second carbon. Resumen

Las nuevas construcciones de ácido nucleico interferente de ARN esférico (siRNA-SNA) parecido a una horquilla superan los desafíos de entrega. Este nuevo diseño mejora la estabilidad del siRNA, reduce la toxicidad y mejora la eficiencia de eliminación de genes para aplicaciones terapéuticas.

Sus antecedentes:

  • La aplicación terapéutica de ARN pequeños interferentes (ARNs si) se ve obstaculizada por su escasa estabilidad y entrega.
  • Las arquitecturas de ácido nucleico esférico (SNA) mejoran la estabilidad del siRNA y la absorción celular.
  • Los diseños de siRNA-SNA híbridos existentes sufren de disociación de la hebra guía, lo que limita la entrega duplex de siRNA activo.

Objetivo del estudio:

  • Introducir e investigar una nueva arquitectura de SNA similar a una horquilla para mejorar la entrega de siRNA.
  • Para superar los problemas de disociación de la hebra guía inherentes a los diseños anteriores de siRNA-SNA.
  • Mejorar la estabilidad, la eficiencia de entrega y las capacidades de silenciamiento de genes de las terapias basadas en siRNA.

Principales métodos:

  • Diseño y síntesis de una molécula similar a una horquilla para inmovilizar ambas hebras de siRNA en un núcleo de SNA.
  • Caracterización de la capacidad de carga, la estabilidad y la citotoxicidad del constructo siRNA-SNA parecido a una horquilla.
  • Evaluación in vitro de la eficiencia y durabilidad de la eliminación de genes en comparación con los siRNA-SNA hibridados.

Principales resultados:

  • La arquitectura similar a una horquilla aumentó la capacidad de carga dúplex de siRNA en 4 veces en comparación con los siRNA-SNA hibridados.
  • Los siRNA-SNA parecidos a alfileres demostraron una semivida 6 veces mayor en suero y una reducción de la citotoxicidad.
  • La destrucción de genes mejorada y más duradera se logró con la construcción siRNA-SNA similar a una horquilla.

Conclusiones:

  • La química de inmovilización mejora significativamente la función biológica de las nanopartículas basadas en siRNA.
  • La arquitectura en forma de horquilla representa una construcción de SNA de próxima generación para una mejor entrega terapéutica de siRNA.
  • Este nuevo diseño es prometedor para el avance de las ciencias de la vida y las aplicaciones de investigación médica de las tecnologías de regulación génica.
