Sus antecedentes:

  • La letalidad sintética es una estrategia poderosa para identificar las interacciones genéticas esenciales y los posibles objetivos farmacológicos.
  • La identificación de compuestos que inhiben las vías bacterianas esenciales es crucial para combatir la resistencia a los antibióticos.

Objetivo del estudio:

  • Aprovechar la letalidad sintética para el descubrimiento de nuevos inhibidores de la biosíntesis de ácido lipoteicoico (LTA) de Staphylococcus aureus.
  • Identificar compuestos dirigidos a la glucosiltransferasa UgtP, esencial para el ensamblaje de LTA.

Principales métodos:

  • Se realizaron pruebas de crecimiento diferencial de aproximadamente 230.000 compuestos.
  • Se utilizaron principios de letalidad sintética para identificar compuestos que inhiben selectivamente las proteínas en vías esenciales.
  • Actividad analizada del compuesto contra el UgtP y su papel en la biosíntesis de LTA.

Principales resultados:

  • Identificaron dos nuevos compuestos como los únicos inhibidores de la biosíntesis de LTA de la biblioteca examinada.
  • Se encontró que ambos compuestos inhiben la UgtP glucosiltransferasa.
  • Los inhibidores restauraron la sensibilidad a la oxacilina en una cepa de Staphylococcus aureus (MRSA) altamente resistente a la meticilina.

Conclusiones:

  • Las pantallas que explotan la letalidad sintética son altamente eficientes para descubrir inhibidores específicos de la vía.
  • Los inhibidores de UgtP identificados representan una nueva vía prometedora para combatir las infecciones por MRSA.
  • Este enfoque es ampliamente aplicable al descubrimiento de fármacos bacterianos y eucariotas.
