Objetivo del estudio:

  • Investigar la interconexión entre la microbiota pulmonar y la reactividad inmune del cerebro.
  • Determinar cómo la desregulación del microbioma pulmonar afecta la susceptibilidad a las enfermedades autoinmunes del sistema nervioso central.
  • Aclarar los mecanismos subyacentes al eje pulmón-cerebro en el desarrollo de enfermedades autoinmunes.

Principales métodos:

  • Se utilizaron modelos de ratas para estudiar el impacto de la manipulación del microbioma pulmonar en el desarrollo de enfermedades autoinmunes.
  • Antibióticos administrados (neomicina, polimicina B) para alterar la composición de la microbiota pulmonar.
  • Evaluación de la activación de las células inmunes, perfiles de citoquinas y signos clínicos de enfermedad autoinmune del sistema nervioso central.

Principales resultados:

  • La desregulación del microbioma pulmonar alteró significativamente la susceptibilidad a la encefalomielitis autoinmune experimental en ratas.
  • El tratamiento con neomicina, que enriquece las bacterias productoras de lipopolisacáridos, indujo un estado de interferón tipo I en la microglía, deteriorando su respuesta a los interferones tipo II.
  • Esto resultó en una reducción de la neuroinflamación, la infiltración de células inmunes y los síntomas clínicos de la enfermedad.

Conclusiones:

  • Existe un eje pulmonar-cerebral funcional, mediado por el microbioma pulmonar.
  • El microbioma pulmonar juega un papel crítico en la regulación de las respuestas inmunes del sistema nervioso central.
  • La modulación de la microbiota pulmonar puede influir en la susceptibilidad y la gravedad de las enfermedades autoinmunes que afectan al cerebro.
