Resumen

La proteína de unión Z-ADN 1 (ZBP1) media el golpe de calor fatal iniciando la muerte celular dependiente de RIPK3. El bloqueo de ZBP1 o las vías de muerte celular relacionadas protege contra el estrés por calor, revelando un nuevo papel para ZBP1 en la termorregulación.

Sus antecedentes:

  • El golpe de calor es una condición crítica relacionada con el estrés por calor, la insuficiencia circulatoria y la disfunción de los órganos.
  • Los mecanismos patógenos del golpe de calor siguen siendo poco conocidos, lo que plantea una creciente preocupación para la salud mundial debido al cambio climático.

Objetivo del estudio:

  • Para aclarar los mecanismos moleculares subyacentes a la patogénesis del golpe de calor.
  • Investigar el papel de la proteína de unión Z-ADN 1 (ZBP1) en la mediación del golpe de calor.

Principales métodos:

  • Se utilizaron modelos de ratón sin ZBP1, RIPK3, MLKL y caspase-8.
  • Análisis de los cambios en la expresión génica y las vías de muerte celular bajo condiciones de estrés por calor.

Principales resultados:

  • El estrés térmico regula la expresión de ZBP1 a través del factor de transcripción de choque térmico 1 (HSF1).
  • ZBP1 activa la muerte celular dependiente de RIPK3, independientemente de la detección del ácido nucleico.
  • La eliminación de ZBP1, RIPK3 o MLKL/caspase-8 redujo significativamente la mortalidad inducida por golpe de calor, la lesión de órganos y la insuficiencia circulatoria.

Conclusiones:

  • ZBP1 juega un papel crítico en la mediación de la patogénesis del golpe de calor a través de la muerte celular dependiente de RIPK3.
  • ZBP1 tiene una función no reconocida anteriormente en la orquestación de las respuestas del huésped al estrés por calor.
  • Dirigirse a ZBP1 y las vías de muerte celular asociadas ofrece estrategias terapéuticas potenciales para el golpe de calor.
