Sus antecedentes:

Objetivo del estudio:

  • Identificar los genes que pueden ser dirigidos para prevenir la disfunción de las células T utilizando pantallas CRISPR de todo el genoma en condiciones inmunosupresoras.
  • Investigar el papel de RASA2 como punto de control de señalización en células T humanas y su potencial como objetivo terapéutico.

Principales métodos:

  • Realizó múltiples pruebas de eliminación de CRISPR en todo el genoma bajo diversas condiciones inmunosupresoras.
  • Investigó el papel de RASA2 en la señalización y función de las células T, incluida la señalización de MAPK y la actividad de las células T CAR.
  • Se evaluó la eficacia de la ablación de RASA2 en modelos preclínicos de terapias con receptores de células T y células T CAR.

Principales resultados:

  • Las pantallas convergieron en RASA2, una proteína activadora de la GTPasa RAS (RasGAP), identificada como un punto de control de señalización de las células T.
  • La ablación de RASA2 mejoró la señalización de MAPK y la actividad citolítica de las células T del receptor de antígeno quimérico (CAR).
  • Las células T con deficiencia de RASA2 mostraron un aumento de la activación, la producción de citoquinas, la actividad metabólica y una mayor matanza de las células cancerosas persistentes.
  • Las células CAR T con knockout de RASA2 mostraron una ventaja de aptitud competitiva in vivo, y la ablación de RASA2 prolongó la supervivencia en modelos preclínicos.

Conclusiones:

  • RASA2 es un objetivo prometedor para mejorar la persistencia de las células T y la función efectiva en el tratamiento del cáncer.
  • La orientación a RASA2 puede superar la disfunción de las células T y mejorar los resultados en las terapias de células T adoptivas.
  • Las estrategias de edición de genes dirigidas a RASA2 tienen potencial para desarrollar tratamientos contra el cáncer más efectivos.
