  • La duración y la actividad de las células GC B (BGC) son críticas para la generación de anticuerpos de alta afinidad, especialmente contra antígenos complejos como el VIH Env.
  • Comprender la dinámica de GC a largo plazo es esencial para diseñar vacunas efectivas contra patógenos desafiantes.

Objetivo del estudio:

  • Investigar la longevidad y las características funcionales de las células B del centro germinal (BGC) después de una inmunización primaria prolongada con la proteína Env del VIH en monos rhesus.
  • Evaluar el impacto de la actividad extendida de GC en la hipermutación somática de anticuerpos, el reconocimiento de epítopos y las respuestas de neutralización de anticuerpos.
  • Evaluar el potencial de una estrategia de inmunización de larga duración y entrega lenta para superar los desafíos de inmunodominancia en el desarrollo de vacunas.

Principales métodos:

  • Los monos rhesus fueron alimentados con la proteína Env del VIH y monitoreados durante hasta 29 semanas sin exposición adicional al antígeno.
  • Se empleó perfiles de transcripción unicelular para analizar los estados de las células GC B (BGC).
  • La hipermutación somática de anticuerpos y las filogenias del linaje celular BGC fueron analizadas para rastrear la evolución y selección de células B.

Principales resultados:

  • Se observaron poblaciones sostenidas de células B del centro germinal (BGC) durante al menos 6 meses, con un aumento significativo de las células BGC en la semana 10.
  • El perfilado de una sola célula confirmó el mantenimiento de estados de GC tanto en la zona clara como en la oscura, con hipermutación y selección somática de anticuerpos en curso.
  • Las células BGC de unión a Env se mantuvieron elevadas a las 29 semanas, y una sola vacunación de refuerzo provocó altos títulos de anticuerpos neutralizantes del VIH.
  • Las células B de memoria generadas en condiciones de larga duración mostraron una mayor hipermutación somática y epítopos no inmunodominantes reconocidos.

Conclusiones:

  • La actividad prolongada de GC, sostenida durante más de 6 meses sin re-inmunización, puede lograrse mediante un enfoque de inmunización de larga duración.
  • Esta fase extendida de GC promueve la evolución significativa de anticuerpos, lo que lleva a potentes respuestas de anticuerpos neutralizantes contra antígenos complejos.
  • La estrategia de inmunización de larga duración y entrega lenta es prometedora para el desarrollo de vacunas contra objetivos difíciles mediante la optimización del ajuste de GC para respuestas máximas de anticuerpos.
