Sus antecedentes:

  • La vía OX40 es crítica para la diferenciación y la memoria de las células T.
  • Rocatinlimab es un anticuerpo anti-OX40 que inhibe esta vía.
  • La dermatitis atópica es una enfermedad inflamatoria crónica de la piel.

Objetivo del estudio:

  • Evaluar la eficacia y seguridad de rocatinlimab en adultos con dermatitis atópica de moderada a grave.
  • Evaluar el impacto de rocatinlimab en las puntuaciones del Índice de Área y Gravedad del Eczema (EASI).
  • Determinar la tolerabilidad y el perfil de eventos adversos de rocatinlimab.

Principales métodos:

  • Un estudio de fase 2b multicéntrico, doble ciego y controlado con placebo.
  • Se incluyeron 274 pacientes adultos con dermatitis atópica de moderada a grave.
  • Los pacientes recibieron rocatinlimab por vía subcutánea (150 mg o 600 mg cada 4 semanas; 300 mg o 600 mg cada 2 semanas) o placebo durante 18 semanas.

Principales resultados:

  • Todos los grupos de rocatinlimab mostraron reducciones significativas del porcentaje medio de mínimos cuadrados en la puntuación EASI a la semana 16 en comparación con el placebo.
  • Las reducciones oscilaron entre -48,3% y -61,1% en todos los brazos de tratamiento con rocatinlimab.
  • Las reacciones adversas comunes incluyeron pirexia, nasofaringitis y escalofríos; no se notificaron muertes.

Conclusiones:

  • El tratamiento con Rocatinlimab condujo a mejoras progresivas en la dermatitis atópica.
  • Estas mejoras se mantuvieron en gran medida después de la interrupción del tratamiento.
  • Rocatinlimab fue bien tolerado en la población del estudio.
