Fusion of Secretory Vesicles with the Plasma Membrane

Proteins and neurotransmitters in secretory vesicles can be released from a cell upon vesicle docking, priming, and fusion with the plasma membrane. Vesicles are docked and primed in preparation for the quick exocytosis of their contents in response to a stimulus. The fusion process is mainly carried out by a SNAP Receptor or SNARE complex, consisting of synaptobrevin, syntaxin-1, and SNAP-25.

  • Para lograr la observación en tiempo real de una sola molécula de la dimerización de las tirosinacinasas receptoras (RTK).
  • Permitir una regulación precisa y un seguimiento dinámico del proceso de dimerización.

Principales métodos:

  • Se utilizaron reglas de plasmones para la observación de las RTK a nivel de una sola molécula.
  • Programación de ADN empleada para la regulación y el monitoreo precisos.
  • Desarrollado nanosondas con excelente fotostabilidad.

Principales resultados:

  • Logró la primera observación en tiempo real de una sola molécula de la dimerización de RTK.
  • Aplicación demostrada de las nanosondas en la regulación de la dimerización, la fosforilación y la señalización de las RTK.
  • Mostró un control preciso sobre los eventos de enlace cruzado de los receptores.

Conclusiones:

  • Las nanosondas propuestas ofrecen un nuevo paradigma para la manipulación precisa y el monitoreo del enlace cruzado de un solo receptor.
  • Estas herramientas pueden dilucidar los mecanismos moleculares de la dimerización y los cambios conformacionales.
  • Potencial significativo para avanzar en la comprensión de las vías de señalización celular.
