Principales métodos:

  • Intercambió dominios de aciltransferasa (AT) en las poliquetases modulares (PKS) para alterar la especificidad del sustrato extensor.
  • Reacciones de biosíntesis de policetida realizadas in vitro utilizando PKS de tipo salvaje y de ingeniería con 14 sustratos extensores diferentes.
  • Desarrolló un flujo de trabajo computacional para predecir el rango de sustrato AT basado en volúmenes de sitio activos.

Principales resultados:

  • Se generaron 13 policetidos estructuralmente distintos, incluidos varios compuestos nuevos, a partir de aproximadamente 200 reacciones in vitro.
  • Las PKS diseñadas produjeron policetidas objetivo más de 100 veces más en algunos casos en comparación con la PKS de tipo salvaje.
  • Los dominios AT inusuales incorporaron preferentemente sustratos extensores raros de tamaño similar o ligeramente más grandes que los sustratos naturales, en lugar de malonilo-CoA o metilmalonilo-CoA.

Conclusiones:

  • El intercambio de dominio de aciltransferasa (AT) es una estrategia viable para la ingeniería de PKS para producir nuevos policetidos in vitro.
  • La comprensión de la especificidad del sustrato de los dominios AT raros amplía la caja de herramientas para la biosíntesis de policetidos.
  • El flujo de trabajo computacional desarrollado ayuda a seleccionar los dominios AT apropiados para la ingeniería PKS y la síntesis química novedosa.
