Sus antecedentes:

  • La visualización de ARN mensajero (ARNm) es un método poderoso para descubrir los ligandos de péptidos de alta afinidad.
  • Las composiciones químicas de ciclización limitada son compatibles con la visualización de ARNm, lo que restringe el descubrimiento de péptidos macrocíclicos.
  • La enzima tirosinasa cataliza la oxidación de la tirosina a una o-quinona reactiva.

Objetivo del estudio:

  • Introducir y caracterizar una nueva ciclización de péptidos mediada por la tirosinasa compatible con la visualización de ARNm.
  • Aplicar este método para descubrir los ligandos macrocíclicos dirigidos al antígeno A4 asociado al melanoma (MAGE-A4).

Principales métodos:

  • Los péptidos que contienen residuos de tirosina y cisteína fueron tratados con tirosinasa para su ciclización.
  • La ciclización mediada por tirosinasa se caracterizó por su aplicabilidad a varios tamaños y andamios de macrociclos.
  • El método desarrollado se combinó con la visualización de ARNm para detectar ligandos dirigidos a MAGE-A4.

Principales resultados:

  • Tratamiento con tirosinasa de péptidos con ciclo rápido que contienen tirosina y cisteína.
  • El método de ciclización demostró ser versátil en diferentes tamaños y andamios de macrociclos.
  • Los ligandos macrocíclicos descubiertos inhiben potentemente el eje de unión MAGE-A4 con valores de IC50 nanomolares.
  • Los ligandos macrocíclicos demostraron una ventaja significativa sobre los análogos no ciclizados, con valores de IC50 que disminuyeron aproximadamente 40 veces.

Conclusiones:

  • La ciclización mediada por tirosinasa es un método ampliamente aplicable y eficiente para generar péptidos macrocíclicos.
  • La combinación de la ciclización de la tirosinasa con la visualización de ARNm es efectiva para descubrir potentes ligandos macrocíclicos.
  • Los ligandos macrocíclicos descubiertos dirigidos a MAGE-A4 muestran potencial terapéutico para el tratamiento del melanoma.
