Resumen

Las células T invariantes asociadas a la mucosa (MAIT) son disfuncionales en el cáncer de hígado y no logran infiltrarse en los tumores. Dirigirse a los macrófagos asociados al tumor que interactúan con las células MAIT puede restaurar su actividad anticancerígena.

Sus antecedentes:

  • Las células T invariables asociadas a la mucosa (MAIT) son células T innatas abundantes en el hígado humano.
  • Las células MAIT desempeñan un papel en la inmunidad y la inflamación, pero su función en el cáncer de hígado no está clara.

Objetivo del estudio:

  • Investigar el papel y las características de las células MAIT en el carcinoma hepatocelular (HCC).
  • Explorar las interacciones celulares que afectan la función de las células MAIT en el microambiente de HCC.

Principales métodos:

  • Secuenciación de ARN unicelular (scRNA-seq) y citometría de flujo en muestras emparejadas de pacientes con CHC.
  • Imágenes de detección conjunta por indexación (CODEX) para el análisis espacial del microambiente tumoral.
  • Herramientas de aprendizaje automático para analizar las redes de interacción celular.
  • Estudios de cocultivo ex vivo y modelos murinos para perturbar las interacciones célula-célula.

Principales resultados:

  • Las células MAIT en HCC presentan heterogeneidad y disfuncionalidad, con una infiltración deteriorada en los tumores hepáticos.
  • Se identificó una interacción específica entre las células MAIT y los macrófagos asociados al tumor CSF1R+PD-L1+ (TAM) como un factor clave en la disfunción de las células MAIT.
  • La alteración de esta interacción de células TAM-MAIT restauró la citotoxicidad de las células MAIT en modelos experimentales.

Conclusiones:

  • La disfunción de las células MAIT es una característica del CHC, influenciada por las interacciones con TAMs específicos.
  • Dirigirse a la interacción entre las células TAM y las MAIT puede mejorar la actividad antitumoral de las células MAIT.
  • Estos hallazgos sugieren que las terapias dirigidas a PD-1/PD-L1 pueden afectar a las células MAIT en pacientes con CHC.
