Sus antecedentes:

  • El mapeo de los receptores de la superficie celular es crucial para el tratamiento de enfermedades como el cáncer, pero presenta desafíos significativos.
  • El control de las interacciones de los receptores es clave para las terapias dirigidas.

Objetivo del estudio:

  • Diseñar nanoestructuras de ADN basadas en aptameros multivalentes ajustables para el control preciso de las interacciones de los receptores de las células tumorales.
  • Desarrollar sistemas programables para mejorar la administración de fármacos dirigidos y la depuración inmune.

Principales métodos:

  • Perfilar los receptores de superficie en 12 líneas celulares utilizando 10 aptamers para crear un mapa de calor distintivo.
  • Incorporación de aptameros en estructuras de origami de ADN (como parches y tubos) para regular el reconocimiento de los receptores.
  • Investigar el reconocimiento mediado por multiheterorreceptores y su efecto sobre la absorción celular y las interacciones inmunes.

Principales resultados:

  • Un mapa de calor distinguía con precisión los tipos de tumor basados en múltiples marcadores de superficie.
  • Las nanoestructuras tubulares aumentaron la absorción intracelular a través de la endocitosis dependiente de la clatrina (> 5 veces el aumento).
  • Las nanoestructuras parecidas a parches promovieron las interacciones de células macrófagas-tumorales para el aclaramiento inmune.

Conclusiones:

  • Las nanoestructuras de ADN multivalentes programables ofrecen una regulación precisa del reconocimiento celular.
  • Este sistema demuestra un potencial significativo para las estrategias personalizadas de tratamiento de tumores.
  • La tecnología facilita la administración de fármacos dirigidos y mejora la inmunidad antitumoral.
