La ferroposis, un proceso de muerte celular, está regulada por la biosíntesis del colesterol. El 7-dehidrocolesterol (7-DHC) actúa como un metabolito antiferroptosis, ofreciendo potencial terapéutico para el cáncer y la lesión por isquemia-reperfusión.

Sus antecedentes:

  • La ferroptosis, una muerte celular dependiente del hierro, está relacionada con enfermedades como el cáncer y la lesión por isquemia-reperfusión (IRI).
  • Las vías de biosíntesis del colesterol son cruciales para la función e integridad celular.

Objetivo del estudio:

  • Investigar el papel de las enzimas distales de la biosíntesis del colesterol en la regulación de la ferroposis.
  • Identificar metabolitos y vías clave involucradas en el control de la ferroposis.

Principales métodos:

  • Cribado CRISPR-Cas9 de todo el genoma para identificar los genes que regulan la ferroposis.
  • Análisis del metabolito para cuantificar los niveles de 7-dehidrocolesterol (7-DHC).
  • Inhibición farmacológica y activación de enzimas específicas en la vía de la biosíntesis del colesterol.

Principales resultados:

  • Las enzimas MSMO1, CYP51A1, EBP y SC5D suprimen la ferroposis, mientras que DHCR7 la promueve.

  • La inhibición de EBP induce la ferroposis y reduce el crecimiento tumoral; la inhibición de DHCR7 promueve la metástasis del cáncer y atenúa la IRI renal.

Conclusiones:

  • El 7-dehidrocolesterol (7-DHC) es un metabolito natural contra la ferroposis.
  • Dirigirse a la vía de biosíntesis de 7-DHC ofrece una estrategia terapéutica prometedora para el cáncer y la IRI.
