Sus antecedentes:

  • Los polisacáridos capsulares (CPS) son componentes cruciales de las envolturas celulares bacterianas, que contribuyen tanto a las interacciones comensales como a las patógenas.
  • La biosíntesis de CPS implica una vía dependiente del transportador ABC, con secreción a través de la envoltura celular mediada por el complejo transportador KpsMT.

Objetivo del estudio:

  • Para dilucidar los mecanismos moleculares subyacentes a la secreción de polisacáridos en las bacterias Gram negativas.
  • Determinar las funciones estructurales y funcionales de los componentes KpsMT, KpsE y KpsD en el transporte CPS.

Principales métodos:

  • Microscopía criolectrónica (Cryo-EM) para determinar la estructura del complejo KpsMT-KpsE en varios estados.
  • Microscopía de fluorescencia de súper resolución para analizar la organización y localización de la superficie celular de CPS.

Conclusiones:

  • Se ha propuesto un modelo molecular detallado para la secreción de polisacáridos en cápsulas bacterianas a través de transportadores ABC.
  • Los hallazgos proporcionan información sobre la base estructural del transporte de CPS y su importancia funcional en la biogénesis de la envoltura celular bacteriana.
