Sus antecedentes:

  • Los órganos de los vertebrados poseen vasos sanguíneos únicos adaptados a sus funciones específicas.
  • El desarrollo de estos vasos específicos de órganos a menudo se considera independiente del proceso de vascularización general.

Objetivo del estudio:

  • Investigar los mecanismos moleculares subyacentes a la formación de vasos sanguíneos específicos del cerebro (angiogénesis).
  • Desafiar la noción de que la especialización organotípica de los vasos no está relacionada con los procesos de vascularización.

Principales métodos:

  • Utilizó la mutagénesis CRISPR-Cas9 en peces cebra para estudiar la función génica.
  • Investigó el papel de los ligandos Wnt7a/b y Mmp25 en la angiogénesis cerebral.
  • Analizó la escisión de las cadenas de colágeno IV por Mmp25.
  • Examinó el impacto de la interferencia genética con la membrana del sótano de pial.

Principales resultados:

  • Se identificó una vía dependiente de Wnt7a/b que controla la angiogénesis cerebral a través de la expresión de Mmp25 en las células endoteliales.
  • Demostró el papel crucial de Mmp25 en la migración de las células de la punta endotelial a través de la membrana del sótano de la piel.
  • Se muestra que el Mmp25 rompe cadenas específicas de colágeno IV, lo que facilita la invasión cerebral.
  • Descubrió que la ruptura de la membrana basal de pial afecta la angiogénesis dependiente de Wnt-β-catenina, lo que lleva a barreras hematoencefálicas defectuosas.

Conclusiones:

  • Se reveló un nuevo mecanismo para la angiogénesis específica de órganos controlada por señales locales (Wnt7a/b) y proteasas específicas (Mmp25).
  • Destacó la importancia de las interacciones de las células de la punta endotelial con la matriz extracelular para la vascularización cerebral.
  • Se estableció un vínculo entre el control organotípico, la angiogénesis y la integridad de la barrera hematoencefálica.
