Sus antecedentes:

  • La difusión intracelular de proteínas cargadas positivamente se suprime, lo que sugiere un papel para la atracción electrostática.

Objetivo del estudio:

  • Para cuantificar cómo se modula la difusión de proteínas por los reactores de carga opuesta y similar en soluciones acuosas.
  • Investigar el papel de la atracción electrostática en las interacciones proteína-proteína no específicas.

Principales métodos:

  • Para medir la difusión de proteínas se utilizaron detección y estadísticas de una sola molécula.
  • Los estados de carga neta de las proteínas se alteraron por la variación del pH, la fuerza iónica y la modificación química.

Principales resultados:

  • Los interactores de carga opuesta suprimieron la difusión a niveles de partes por millón.
  • Los interactores de la misma carga no afectaron la difusión más allá de la concentración del 1%.
  • La supresión de la difusión fue sensible a la carga neta de proteínas y robusta en diferentes pares de proteínas.

Conclusiones:

  • Existen interacciones proteína-proteína ubicuas, impulsadas por la carga neta.
  • La atracción electrostática juega un papel clave en la supresión de la difusión, ofreciendo información sobre los mecanismos celulares.
