  • Evaluar su citotoxicidad, absorción celular y mecanismo de acción.
  • Para evaluar su toxicidad preliminar en un modelo animal.

Principales métodos:

  • Cribado de citotoxicidad en varias líneas celulares.
  • Estudios de absorción celular mediante la luminiscencia intrínseca.
  • Microscopía confocal y nanoscopía de agotamiento de emisión estimulada (STED).

  • Ambos cationes mostraron una citotoxicidad selectiva contra las células cancerosas frente a las células HEK293.

  • Los estudios de absorción celular revelaron una internalización preferencial del catión más lipofílico.
  • El tratamiento dio lugar a hinchazón lisosómica, disfunción mitocondrial y muerte celular dependiente del lisosoma (necrosis y apoptosis).
  • No se observó toxicidad detectable en el modelo de Galleria melonella hasta 80 mg/kg.

Conclusiones:

  • Los cationes orgánicos polipirídicos representan una clase prometedora de compuestos fotoactivos para el desarrollo terapéutico.
  • El catión activo demuestra una potente citotoxicidad activada por la luz con un perfil de seguridad preliminar favorable.
  • Se justifica una investigación adicional para explorar su eficacia terapéutica y optimizar su diseño.
