Sus antecedentes:

  • El acortamiento de los telómeros y la reducción de la actividad de la transcriptasa inversa de la telomerasa (TERT) están relacionados con el envejecimiento y las enfermedades relacionadas con la edad.
  • El TERT juega un doble papel en el mantenimiento de los telómeros y como co-regulador de la transcripción en los procesos de envejecimiento.

Objetivo del estudio:

  • Para identificar y caracterizar un compuesto que activa la transcripción TERT.
  • Evaluar el potencial terapéutico de la activación de TERT en la mitigación de las características del envejecimiento y las patologías asociadas.

Principales métodos:

  • Identificación de un compuesto activador de TERT (TAC) que modula la vía de señalización MEK/ERK/AP-1.
  • Evaluación de los efectos del TAC sobre los niveles de TERT, la síntesis de telómeros, la senescencia celular y los marcadores inflamatorios en células humanas y ratones envejecidos.
  • Evaluación del impacto del TAC en la neuroinflamación, la neurogénesis y la función cognitiva in vivo.

Principales resultados:

  • TAC regula con éxito la transcripción de TERT y la síntesis de telómeros en células humanas primarias y ratones envejecidos.
  • El tratamiento con TAC redujo la senescencia celular, las citoquinas inflamatorias y silenció la expresión de p16INK4a a través de la hipermetilación mediada por DNMT3B.
  • En el cerebro, el TAC alivió la neuroinflamación, mejoró los factores neurotróficos, estimuló la neurogénesis y preservó la función cognitiva sin inducir toxicidad ni aumentar el riesgo de cáncer.

Conclusiones:

  • La activación fisiológica del TERT es una estrategia viable para combatir múltiples señales del envejecimiento.
  • El compuesto activador de TERT identificado muestra una promesa preclínica para tratar enfermedades asociadas a la edad y promover un envejecimiento más saludable.
