Principales métodos:
- Integración sistemática de 25 conjuntos de datos de secuenciación de ARN de una sola célula.
- Procesamiento uniforme de 385 muestras de 189 controles sanos mediante scAutoQC.
- Anclado de 12 conjuntos de datos de enfermedades en el atlas de referencia saludable.
Principales resultados:
- Desarrollo de un atlas de referencia saludable con ~ 1,1 millones de células y 136 estados celulares.
- Descubrimiento de la metaplasia de células epiteliales en enfermedades inflamatorias, parecidas a las células de las glándulas pilóricas y de Brunner.
- Identificación de las células metaplásticas de la glándula pilórica que reclutan células inmunes como las células T y los neutrófilos.
Conclusiones:
- La inflamación induce cambios en las células madre que alteran la arquitectura de la mucosa y promueven una mayor inflamación.
