Sus antecedentes:

  • El zinc es un micronutriente vital que regula los procesos fisiológicos, principalmente a través de la unión a los residuos de proteína cisteína.
  • Los sitios específicos de cisteína involucrados en la unión de zinc a través del proteoma humano no están caracterizados en gran medida.
  • Comprender las interacciones zinc-proteína es crucial para modular la función de las proteínas y los procesos celulares.

Objetivo del estudio:

  • Desarrollar un mapa completo y cuantitativo del proteoma de cisteína de unión al zinc (ZnCPT).
  • Identificar y caracterizar nuevos sitios de unión al zinc de la cisteína dentro de las proteínas humanas.
  • Explorar las implicaciones funcionales de la unión de zinc en diversos dominios biológicos y contextos de enfermedades.

Principales métodos:

  • Desarrollo de ZnCPT, una técnica de mapeo proteómico profundo y cuantitativo.
  • Identificación sistemática de los residuos de cisteína implicados en la unión de zinc en el proteoma humano.
  • Análisis de las familias de proteínas y de los dominios funcionales regulados por la unión al zinc.

Principales resultados:

  • Se han definido 6.173 cisteínas que se unen al zinc, lo que revela una modulación generalizada de la función de la proteína por zinc.
  • Familias de proteínas identificadas en los principales dominios biológicos afectados por la unión constitutiva o inducible al zinc.
  • Descubrió 52 dependencias genéticas de cáncer influenciadas por la unión de zinc, incluidos los cánceres de pulmón dependientes de GSR.

Conclusiones:

  • El ZnCPT proporciona un recurso valioso para comprender la regulación del zinc de la función de las proteínas.
  • La unión al zinc juega un papel importante en la regulación estructural, enzimática y alosterica de las proteínas.
  • Los hallazgos nombran tumores malignos sensibles a la citotoxicidad inducida por el zinc y revelan una nueva interacción zinc-GSR en el cáncer de pulmón.
