Sus antecedentes:

  • Las desmetilasas de lisina (KDM) son reguladores cruciales de la expresión génica, utilizando hierro y 2-oxoglutarato (2OG).
  • Los KDM son objetivos farmacológicos importantes para la terapia del cáncer, pero su diversidad funcional requiere sondas químicas precisas.
  • La comprensión mecanicista limitada de la heterogeneidad de la DMC humana dificulta el desarrollo terapéutico dirigido.

Objetivo del estudio:

  • Desarrollar mutantes sensibles al análogo de miembros de la subfamilia KDM4 para diseccionar funciones biológicas individuales.
  • Crear sondas químicas para la inhibición temporal y específica de la actividad de KDM4.
  • Aclarar el papel de miembros específicos de KDM4, en particular KDM4A, en los procesos celulares y el cáncer.

Principales métodos:

  • La ingeniería de los mutantes KDM4 mediante la sustitución de la fenilalanina del sitio activo por la alanina, creando variantes sensibles al análogo.
  • Desarrollo y aplicación de inhibidores competitivos de los cofactores, como la N-oxalil leucina (NOL), dirigidos a los as-mutantes.

  • Se generaron con éxito KDM4 como mutantes con actividad catalítica de tipo salvaje y especificidad de sustrato.
  • Se ha identificado a la N-oxalil leucina (NOL) como un inhibidor potente y reversible de los KDM4 como mutantes con eficacia submicromolar.
  • Capacidad demostrada de los prodrogas de NOL para modular la metilación de la histona lisina en células humanas cultivadas.
  • Se descubrió una función específica de KDM4A en la promoción de la síntesis de proteínas ribosómicas a través de la desmetilación de histonas específicas del locus.

Conclusiones:

  • Los mutantes KDM4 sensibles al análogo y los inhibidores NOL proporcionan herramientas poderosas para diseccionar la heterogeneidad funcional de KDM.
  • KDM4A juega un papel crítico en la regulación de la expresión de rRNA, el ensamblaje de ribosomas y la síntesis de proteínas.
